Rhine McLin, then a state senator, gave Nina her first professional job as a legislative aide. She then returned to her hometown to join the administration of Mayor Michael White, who promoted her to Executive Assistant for Legislative Affairs. As Director of Government Affairs for the Cleveland Metropolitan School District, she lobbied on behalf of Cleveland school children at both the federal and state levels.

In 2001, she ran for office in Cleveland City Council Representative for Ward 1, but incumbent Joe Jones was re-elected, although he resigned three years later in November 2004. His wife, Tonya Jones, returned to the race in the 2005 election, but was defeated by Nina, who took office. as a representative of Ward One Town Council.

Nina Turner replaced Senator Lance Mason who resigned from his 25th district in the Ohio Senate in favor of appointment to the Cuyahoga County Court of Common Pleas in September 2008.
